UV), high hydrostatic pressure, fermentation or the addition of antimicrobials and chemicals, can induce different physiological states in bacteria, and increase the possibility of inducing the transition to survival mechanisms including spore formation or the <strong>transition for non-spore-forming microorganisms to viable but not culturable cells (VBNC)<\/strong>.<\/p>\n<p><img fetchpriority=\"high\" decoding=\"async\" class=\"size-full wp-image-11191 aligncenter\" src=\"https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-VBNC-Viable-but-not-colturable-cell.jpg\" alt=\"Piramide VBNC - Viable but not colturable cell\" width=\"548\" height=\"528\" srcset=\"https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-VBNC-Viable-but-not-colturable-cell.jpg 548w, https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-VBNC-Viable-but-not-colturable-cell-300x289.jpg 300w\" sizes=\"(max-width: 548px) 100vw, 548px\" \/><\/p>\n<p>To date, more than 60 bacterial species capable of entering the VBNC state have been identified, including Escherichia, Vibrio, Listeria and Pseudomonas.<\/p>\n<p><img decoding=\"async\" class=\"size-full wp-image-11192 aligncenter\" src=\"https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-VBNC-Patogeni.jpg\" alt=\"Piramide VBNC - Patogeni\" width=\"1218\" height=\"506\" srcset=\"https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-VBNC-Patogeni.jpg 1218w, https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-VBNC-Patogeni-300x125.jpg 300w, https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-VBNC-Patogeni-1024x425.jpg 1024w, https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-VBNC-Patogeni-768x319.jpg 768w\" sizes=\"(max-width: 1218px) 100vw, 1218px\" \/><\/p>\n\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-102548a8 elementor-widget__width-initial elementor-widget elementor-widget-text-editor\" data-id=\"102548a8\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<h2><strong>VBNC cell properties<\/strong><\/h2>\n<p><img decoding=\"async\" class=\"alignnone size-full wp-image-11201\" src=\"https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-Proprieta-cellule-VBNC.jpg\" alt=\"Piramide - Propriet\u00e0 cellule VBNC\" width=\"900\" height=\"300\" srcset=\"https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-Proprieta-cellule-VBNC.jpg 900w, https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-Proprieta-cellule-VBNC-300x100.jpg 300w, https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-Proprieta-cellule-VBNC-768x256.jpg 768w\" sizes=\"(max-width: 900px) 100vw, 900px\" \/><\/p>\n<p>VBNC cells have some specific characteristics.<\/p>\n<p>Compared to dead cells, VBNC cells have an intact membrane, successfully maintain gene expression and avoid cytoplasmic leakage.<\/p>\n<p>In contrast, dead cells have a damaged membrane and are metabolically inactive.<\/p>\n<p>Similarly, compared to viable and sub-lethally damaged cells, VBNC cells cannot grow in the standard medium, which is a specific character of VBNC cells. Furthermore, physiological and molecular differences exist between VBNC cells and viable and culturable cells, including cell wall and membrane compositions, gene expression, adhesion properties, virulence potential, metabolism, cell morphology, and physical and chemical resistance.<\/p>\n<p>For instance, cell size reduction was found in VBNC E. coli, while cell dwarfing and round cells were described for some VBNC cells. Furthermore, VBNC cells have a low metabolism and a high ATP level. However, the mechanism by which bacteria adapt to the VBNC state remains unclear.<\/p>\n<p>The ability to return to the viable state is also an advantageous feature of VBNC cells, and in the scientific literature this process is described as \u2018resuscitation\u2019. Resuscitation is a very important feature of VBNC bacteria. <u>VBNC cells that respond to appropriate environmental stimuli, such as an increase in temperature or the addition of nutrients, become metabolically active and culturable again.<\/u><\/p>\n<p><u>This implies that appropriate conditions could be provided during food processing for the resurrection of VBNC bacteria, leading to a risk in terms of food quality and safety.<\/u> Indeed, many pathogenic bacteria in the VBNC state are not able to initiate infection, but retain virulence potential, which will only be expressed following resuscitation.<\/p>\n<p>This is why their control poses a great challenge to the food industry.<\/p>\n\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-2a58fef elementor-widget__width-initial elementor-widget elementor-widget-text-editor\" data-id=\"2a58fef\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<h2><strong>Identification of VBNC cells<\/strong><\/h2>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-11202\" src=\"https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-Identicazione-cellule-VBNC.jpg\" alt=\"Piramide - Identicazione cellule VBNC\" width=\"900\" height=\"300\" srcset=\"https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-Identicazione-cellule-VBNC.jpg 900w, https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-Identicazione-cellule-VBNC-300x100.jpg 300w, https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-Identicazione-cellule-VBNC-768x256.jpg 768w\" sizes=\"(max-width: 900px) 100vw, 900px\" \/><\/p>\n<p><u>In the VBNC state, bacteria cannot grow on conventional media and can therefore evade conventional detection methods.<\/u><\/p>\n<p>In fact, micro-organisms in the non-culturable state, not growing on the routine bacteriological media on which they would normally grow and develop into colonies, can cause numerous false negatives. An example is given below for E. coli, where the presence of VBNC from E. coli makes its monitoring more difficult and is likely to produce false negative results.<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"size-full wp-image-11196 aligncenter\" src=\"https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-Identificazione-delle-cellule-VBNC.jpg\" alt=\"Piramide - Identificazione delle cellule VBNC\" width=\"460\" height=\"518\" srcset=\"https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-Identificazione-delle-cellule-VBNC.jpg 460w, https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-Identificazione-delle-cellule-VBNC-266x300.jpg 266w\" sizes=\"(max-width: 460px) 100vw, 460px\" \/><\/p>\n<p>As a result, while for a long time it was believed that a bacterial cell died when it was no longer able to grow on normal culture media, we now know that this assumption is simplistic and that there are many situations in which a cell loses viability but remains viable and potentially able to regrow.<br \/>\nBecause of this, the detection of VBNC cells presents a major challenge as they can be the source of a potential food safety and human health risk.<br \/>\nSince cells in the VBNC state are no longer culturable, alternative \u2018non-culture\u2019 methods must be used in order to prove that cells in this state are alive. The table below lists some of the methods reported in the literature that can be used to detect VBNC cells. These are complemented by even more modern approaches such as metagenomics, which combines PCR with bioinformatic analysis, and MALDI-TOF technology.<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-11197\" style=\"font-style: inherit; font-weight: inherit;\" src=\"https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-metodi-per-rilevare-cellule-VBNC.jpg\" alt=\"Piramide - metodi per rilevare cellule VBNC\" width=\"873\" height=\"395\" srcset=\"https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-metodi-per-rilevare-cellule-VBNC.jpg 873w, https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-metodi-per-rilevare-cellule-VBNC-300x136.jpg 300w, https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-metodi-per-rilevare-cellule-VBNC-768x347.jpg 768w\" sizes=\"(max-width: 873px) 100vw, 873px\" \/><\/p>\n<p>Advances in molecular technologies have made it possible to understand how traditional microbiology methods are able to estimate less than 1% of the approximately 109 bacterial cells in one gram of medium.<\/p>\n<p>These studies concluded that the standard incubation times used in laboratory experiments were insufficient for the bacterial growth of a number of microbial strains and a large part of this underestimation is due to the presence of non-culturable cells.<\/p>\n<p>The lack of information that exists of the remaining 99% of the microbial population is not insignificant as it can be crucial in understanding various phenomena such as biofilm formation, the emergence of resistance, and quorum sensing (QM), the mechanisms of which have yet to be exactly deciphered in many species.<\/p>\n\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-bcd8c04 elementor-widget__width-initial elementor-widget elementor-widget-text-editor\" data-id=\"bcd8c04\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<h2><strong>The Piramide and Realco approach<\/strong><\/h2>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-11203\" src=\"https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-e-Realco-Azioni-specifiche-per-contaminazioni-da-VBNC.jpg\" alt=\"Piramide e Realco - Azioni specifiche per contaminazioni da VBNC\" width=\"900\" height=\"300\" srcset=\"https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-e-Realco-Azioni-specifiche-per-contaminazioni-da-VBNC.jpg 900w, https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-e-Realco-Azioni-specifiche-per-contaminazioni-da-VBNC-300x100.jpg 300w, https:\/\/www.piramide-ambiente.it\/wp-content\/uploads\/2023\/07\/Piramide-e-Realco-Azioni-specifiche-per-contaminazioni-da-VBNC-768x256.jpg 768w\" sizes=\"(max-width: 900px) 100vw, 900px\" \/><\/p>\n<p>For the above reasons, it is essential for the food industry (and beyond) to implement specific actions to control biofilm contamination.<\/p>\n<p>To this end, the application of the Biorem\u00ae patent is useful.
