Filtration and packaging lines<\/h4>\n\n<ul class=\"wp-block-list\">\n<li><strong>Filters<\/strong>: Duplex filters located upstream of homogenizers and DSI systems can trap microorganisms that go on to form biofilms.<\/li>\n\n\n\n<li><strong>Packaging machines<\/strong>: These represent a critical point for post-pasteurization contamination.<\/li>\n\n\n\n<li><strong>Spray nozzles<\/strong>: In powder production facilities, even the <em> spray nozzles<\/em> at the end of the process can harbor bacteria.<\/li>\n<\/ul>\n\n<h2 class=\"wp-block-heading\" id=\"h-meccanismi-di-adesione\">Adhesion mechanisms<\/h2>\n\n<p class=\"wp-block-paragraph\">The adhesion of <strong>Bacillus cereus<\/strong> spores to stainless steel surfaces is a complex phenomenon resulting from a combination of structural characteristics, physicochemical properties, and environmental factors.<\/p>\n\n<h3 class=\"wp-block-heading\" id=\"h-le-ragioni-principali-di-questa-tenace-adesione-sono\">The main reasons for this steadfast commitment are:<\/h3>\n\n<ul class=\"wp-block-list\">\n<li><strong>Surface hydrophobicity<\/strong>: Spores possess high <strong>surface hydrophobicity<\/strong>, which facilitates their binding to inert materials such as stainless steel. This physicochemical property is one of the key factors that allows the spore to &#8220;cling&#8221; to the surface. <\/li>\n\n\n\n<li><strong>External cellular structures<\/strong>: Adhesion capacity is closely linked to the presence of specific structures on the spore\u2019s outer surface, particularly <strong>the exosporium<\/strong> (an outer protective membrane) and the appendages (or filaments). These structures act as anchoring organs that facilitate initial contact and subsequent stable bonding with steel. <\/li>\n\n\n\n<li><strong>Genetic and proteomic potential<\/strong>: Adhesion and subsequent growth within the biofilm are supported by the bacterium\u2019s genetic potential. Proteomic studies have shown that, as early as the first two hours of biofilm formation, B. cereus expresses specific proteins that facilitate the colonization process. <\/li>\n\n\n\n<li><strong>Mechanical factors (Flow)<\/strong>: Processing conditions affect adhesion. For example, <strong>turbulent flow<\/strong> within the equipment can promote adhesion by physically bringing the spores closer to the equipment walls than under static conditions. <\/li>\n\n\n\n<li><strong>Resistance to cleaning:<\/strong> Spores not only adhere easily, but are also capable of surviving standard cleaning cycles (CIP). In some cases, chemical treatments can even select for spores with specific surface properties that make them even more effective at adhering.   <\/li>\n<\/ul>\n\n<p class=\"wp-block-paragraph\">Once attached, these spores can germinate, multiply, and form mature biofilms that become chronic reservoirs of contamination, protecting the bacteria from disinfectants and making it difficult to eradicate them from production lines.  <\/p>\n\n<h2 class=\"wp-block-heading\" id=\"h-perche-il-sistema-cip-standard-puo-fallire\">Why can the standard CIP system fail?  <\/h2>\n\n<p class=\"wp-block-paragraph\">The failure of traditional <strong>Cleaning-in-Place (CIP)<\/strong> systems to combat biofilms is due to a combination of structural, chemical, and biological factors that make these microbial communities extremely resilient.<\/p>\n\n<h3 class=\"wp-block-heading\" id=\"h-le-ragioni-principali-per-cui-il-cip-non-riesce-a-eradicare-completamente-questi-biofilm-latte-in-polvere-sono\">The main reasons why CIP is unable to completely remove these milk powder biofilms are:<\/h3>\n\n<div style=\"height:30px\" aria-hidden=\"true\" class=\"wp-block-spacer\"><\/div>\n\n<h4 class=\"wp-block-heading\" id=\"h-1-la-barriera-protettiva-della-matrice-eps\">1 &#8211; The protective barrier of the EPS matrix<\/h4>\n\n<p class=\"wp-block-paragraph\">Biofilms are not simply accumulations of bacteria, but communities embedded in a matrix of extracellular polymeric substances (<a href=\"https:\/\/www.piramide-ambiente.it\/en\/role-of-eps-in-biofilms-part-i\/\"><mark style=\"background-color:rgba(0, 0, 0, 0);color:#00d084\" class=\"has-inline-color\">EPS<\/mark><\/a>) produced by the bacteria themselves. This matrix acts as a physical and chemical shield that: <\/p>\n\n<ul class=\"wp-block-list\">\n<li>It <strong>protects bacteria<\/strong> from the action of biocides, rendering them inaccessible.<\/li>\n\n\n\n<li>It enables the microorganisms within the biofilm to withstand changes in temperature and pH much better than their planktonic counterparts (those floating freely in the milk).<\/li>\n\n\n\n<li>It requires very harsh treatments using powerful oxidizing agents or specific enzymes to be dissolved, which is beyond the capabilities of standard detergents.<\/li>\n<\/ul>\n\n<h4 class=\"wp-block-heading\" id=\"h-2-tolleranza-chimica-e-adattamento\">2 &#8211; This action \u201copens up\u201d the protective structure, allowing disinfectants to more easily reach and kill the bacteria nestled inside. <\/li>\n\n\n\n<li><strong>Incorporation into<\/strong> the <strong>CIP<\/strong> system: The addition of enzymes to the CIP system\u2019s wash solutions has been shown to significantly improve biofilm removal efficiency compared to chemical treatments alone.<\/li>\n\n\n\n<li><strong>Optimization of alkaline washing<\/strong>: Sources suggest enhancing the alkaline washing step with enzymatic treatments to promote more thorough removal of biofilm from surfaces.
